This study investigated the genetic link between recurrent spontaneous abortion (RSA) and specific cytokine gene polymorphisms. Certain tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), and interleukin-10 (IL-10) gene variations are associated with RSA.
Area of Science:
- Immunogenetics
- Reproductive Immunology
- Molecular Genetics
Background:
- Recurrent spontaneous abortion (RSA) affects 3-5% of pregnancies.
- Immunological dysregulation is implicated in RSA pathogenesis.
- Cytokines, as key immune mediators, are potential factors in successful pregnancy.
Purpose of the Study:
- To investigate the association between polymorphisms in pro-inflammatory (TNF-α, TNF-β) and anti-inflammatory (IL-6, IL-10) cytokine genes and RSA.
- To compare serum cytokine levels between women with RSA and healthy controls.
Main Methods:
- Genotyping of TNF-α (-308 G→A, -238 G→A), TNF-β (+252 G→A), IL-6 (-634 G→C, -174 G→C), and IL-10 (-1082 A→G, -819 C→T, -592C→A) polymorphisms using PCR-based methods.
- Quantification of serum TNF-α, TNF-β, IL-6, and IL-10 levels via ELISA.
- Case-control study involving 284 women with RSA and 284 controls.
Main Results:
- Significant differences in TNF-α -308 G/A, IL-6 -174 G/C, and IL-10 -819 G/C polymorphisms were observed between RSA patients and controls (P=0.008, P=0.0005, P=0.03, respectively).
- Serum levels of IL-6 and IL-10 were significantly lower in women with RSA compared to controls (P<0.05).
- No significant differences in serum TNF-α and TNF-β levels were found between groups (P>0.05).
Conclusions:
- The study suggests a potential association between specific promoter polymorphisms in TNF-α (-308), IL-6 (-174), and IL-10 (-819) genes and the risk of recurrent spontaneous abortion.
- Altered expression of IL-6 and IL-10 may play a role in the pathophysiology of RSA.
