Studies on the tissue specificity of a circulating renotropic factor.

    Summary

Sera from nephrectomized rats stimulate kidney cell DNA synthesis. A renal tissue factor appears to activate this renotropic substance, enabling it to act on liver cells as well.

    Area of Science:

    • Nephrology
    • Molecular Biology
    • Biochemistry

    Background:

    • Sera from unilaterally nephrectomized rats stimulate DNA synthesis in renal cortex tissue.
    • This effect was previously considered specific to kidney tissue.
    • A renal tissue factor was identified that enhances the action of circulating renotropic factors.

    Purpose of the Study:

    • To investigate the role of the renal tissue factor in the specificity of serum-stimulated DNA synthesis.
    • To determine if the renal tissue factor can enable non-renal tissues to respond to renotropic factors.

    Main Methods:

    • Incubation of rat renal cortex slices with sera from nephrectomized rats.
    • Incubation of rat liver, spleen, and lung slices with sera from nephrectomized rats.
    • Addition of a renal tissue factor to liver slices incubated with sera from uninephrectomized rats.
    • Measurement of 3H-Thymidine incorporation into DNA as an indicator of cell proliferation.

    Main Results:

    • Sera from nephrectomized rats stimulated 3H-Thymidine incorporation into renal cortex DNA.
    • These sera did not stimulate isotope incorporation in liver, spleen, or lung tissue.
    • The addition of the renal tissue factor and sera from uninephrectomized rats stimulated 3H-Thymidine incorporation into liver slices.

    Conclusions:

    • The renal tissue factor plays a crucial role in activating circulating renotropic substances.
    • This activation mechanism explains the observed specificity of renotropic factors for renal tissue.
    • The renal tissue factor can enable non-renal tissues, such as the liver, to respond to renotropic stimulation.
