    Background:

    • Calcitonin is a crucial hormone involved in calcium regulation.

    Purpose of the Study:

    • To develop a sensitive radioimmunoassay (RIA) for quantifying human calcitonin.
    • To establish normal plasma calcitonin levels in healthy adults.
    • To investigate the effects of various secretagogues on calcitonin secretion.

    Main Methods:

    • Development of a highly sensitive radioimmunoassay (RIA) capable of detecting 1-2 pg of hormone.
    • Measurement of plasma calcitonin concentrations in 55 normal adult subjects.

    • The developed RIA allows for the measurement of low calcitonin concentrations in unextracted human plasma.

    • Calcium, pentagastrin, and glucagon significantly stimulated plasma calcitonin; food and oral calcium did not. Pentagastrin's effect was more pronounced in males.

    Conclusions:

    • The study successfully developed a sensitive RIA for human calcitonin.
    • Basal calcitonin levels show no significant age or sex differences.
    • Specific secretagogues can stimulate calcitonin release, suggesting potential sex-based differences in calcitonin reserve.
