We developed a novel protein microarray biosensor for rapid, simultaneous detection of multiple hepatitis B virus (HBV) markers. This SPR-based platform offers high sensitivity and specificity for efficient HBV diagnosis.
Area of Science:
- Biomedical Engineering
- Immunotechnology
- Viral Diagnostics
Background:
- Hepatitis B virus (HBV) infection is a significant global health concern.
- Current diagnostic methods for HBV serological markers are often time-consuming and detect markers individually.
Purpose of the Study:
- To develop and optimize a protein microarray-based biosensor for simultaneous detection of multiple HBV serological markers.
- To evaluate the performance of the biosensor in terms of sensitivity, specificity, and speed compared to existing methods.
Main Methods:
- Development of a surface plasmon resonance (SPR) based protein microarray.
- Optimization of printing buffer, protein immobilization time, and capture protein concentration.
- Simultaneous detection of five HBV markers in human serum samples.
Main Results:
- The optimized biosensor detected five HBV markers in 20 μL of human serum within 30 minutes.
- Achieved a sensitivity of 0.1 ng/mL for Hepatitis B surface antigen (HBsAg).
- Demonstrated 85% agreement with commercial ELISA kits in analyzing 35 patient samples, showing high specificity, reproducibility, and stability.
Conclusions:
- The integrated protein microarray and SPR technique provides a rapid, sensitive, and specific platform for HBV diagnosis.
- This high-throughput method offers a significant advancement over conventional ELISA and PCR techniques for HBV marker detection.
