Purpose of the Study:

  • To investigate the role of Notch signaling in the cellular differentiation processes within pleomorphic adenoma.
  • To correlate Notch expression patterns with specific cell types and differentiation stages in PA.

Main Methods:

  • Immunohistochemistry was employed to detect Notch expression in 30 cases of pleomorphic adenoma.
  • Expression levels and localization of Notch were analyzed in ductal epithelial cells, solid tumor nests, and areas of squamous metaplasia.

Main Results:

  • Partial CK7 expression and substantial Notch expression were observed in ductal epithelial cells, suggesting Notch involvement in differentiation.
  • Notch expression was detected in the nucleus of basal and squamous cells, with intense expression in basal cells and weak expression in squamous cells, indicating a role in basal to squamous cell differentiation.
  • Loss of nuclear Notch expression in inner layers and its presence in cytoplasm/cell membrane of cartilage/mucous cells suggested the termination of differentiation.

Conclusions:

  • Notch expression patterns in basal cells undergoing squamous metaplasia further support its participation in PA differentiation.
  • These findings highlight Notch as a key player in the differentiation pathways observed in pleomorphic adenoma.
