- N-3PUFA supplementation showed a small to modest benefit in reducing depressive symptoms versus placebo (SMD -0.32), but with very low-quality evidence and potential bias.
- Adverse events, remission, response rates, and quality of life were similar between n-3PUFA and placebo groups, with wide confidence intervals. The single antidepressant comparison showed no significant difference.
Conclusions:
- High-quality evidence to definitively determine the effects of n-3PUFAs for MDD is currently insufficient.
- The observed small benefit of n-3PUFAs over placebo is likely not clinically meaningful and may be overestimated due to low-quality evidence and potential bias.
- Further research with robust methodology is required to establish the true efficacy and safety profile of n-3PUFAs in MDD treatment.
