Enzyme-Linked Immunosorbent Assay01:33

Enzyme-Linked Immunosorbent Assay

18.6K
In 1971, Peter Perlman and Eva Engvall developed an Enzyme-linked immunosorbent assay (ELISA or EIA). ELISA differs from western blot in that the assays are conducted in microtiter plates or in vivo rather than on an absorbent membrane.
There are many different types of ELISAs, but they all involve an antibody molecule whose constant region binds an enzyme, leaving the variable region free to bind its specific antigen.  Enzyme-substrate reaction allows the antigen to be visualized or...

Area of Science:

  • Chemical biology
  • Molecular medicine
  • Biotechnology

Background:

  • Targeting cell surface proteins is crucial but challenging for disease treatment.
  • Aptamers, generated via Systematic Evolution of Ligands by Exponential enrichment (SELEX), are promising ligands.
  • Cell-SELEX adapts SELEX for complex targets like cell surfaces.

Purpose of the Study:

  • To provide an overview of recent advances in cell-SELEX technology.
  • To describe a differential cell-SELEX approach for identifying cancer signatures.
  • To outline methods for evaluating aptamer specificity and identifying target receptors.

Main Methods:

  • Overview of cell-SELEX technology advancements.
  • Detailed description of the differential cell-SELEX protocol.
  • Methods for assessing aptamer binding specificity.
  • Preliminary identification of potential target receptors.

Main Results:

  • The differential cell-SELEX approach enables identification of specific aptamers for cancer cell surfaces.
  • This method has been applied to identify signatures for malignant glioma and non-small-cell lung cancer.
  • Established procedures for evaluating specificity and identifying targets.

Conclusions:

  • Cell-SELEX is an effective strategy for aptamer generation against cell surface targets.
  • Differential cell-SELEX offers a powerful tool for cancer biomarker discovery.
  • This technology holds potential for advancing molecular medicine and diagnostics.
