Area of Science:

  • Oncology
  • Pharmacology
  • Radiation Oncology

Background:

  • 5-Fluorouracil (FUra) is a key radiosensitizer, increasing cellular sensitivity to x-rays.
  • Early research established FUra's radiosensitizing capacity.
  • The critical role of specific dosing schedules for FUra's effectiveness was recently identified.

Purpose of the Study:

  • To determine the optimal administration of 5-Fluorouracil (FUra) for enhanced radiosensitization.
  • To investigate the clinico-pharmacologic requirements for maximizing FUra's radiosensitizing utility.
  • To evaluate the efficacy of infused FUra in combination with radiation therapy for specific cancer types.

Main Methods:

  • Identification of extracellular drug concentration and time factors for intracellular radiosensitization.
  • Conducting clinico-pharmacologic investigations to establish optimal treatment protocols.
  • Implementing prolonged continuous infusions of FUra combined with fractionated, cyclical radiation therapy.

Main Results:

  • Prolonged continuous infusions of FUra with fractionated radiation maximize clinical utility.
  • Infused FUra radiosensitization therapy demonstrates maximum efficacy against squamous-transitional cancers.
  • Preliminary data indicates significant promise for rectal and breast cancer local/regional control.

Conclusions:

  • Optimized FUra infusion protocols combined with radiation therapy offer significant clinical benefits.
  • This approach holds potential to substantially decrease the need for radical cancer surgery.
  • 5-Fluorouracil is a promising radiosensitizer with broad applicability in cancer treatment.
