Formulation and Manufacturing Process: Physical Attributes of Generic Tablets and Capsules01:18

Formulation and Manufacturing Process: Physical Attributes of Generic Tablets and Capsules

403
Bioequivalence in generic drugs, such as tablets and capsules, refers to their pharmaceutical equivalence to the brand-name counterparts. However, for therapeutic equivalence, manufacturers must also consider physical attributes like size, shape, and weight (FDA Guidance for Industry, December 2003). Discrepancies in these aspects could impact patient compliance and cause medication errors. For instance, swallowing difficulties, often experienced with larger tablets or capsules, can lead to...

Factors Influencing Drug Absorption: Pharmaceutical Parameters

761
Solid dosage forms such as tablets and capsules undergo rigorous manufacturing processes to ensure stability and effectiveness. Their dissolution and absorption properties are influenced significantly by the choice of excipients (inactive ingredients that serve various roles in the formulation), and the methodology applied during production. The manufacturing parameters, such as compression force and granulation techniques, significantly affect dissolution rates. Elevated compression forces...

Factors Influencing Drug Absorption: Physicochemical Parameters

1.2K
The physicochemical characteristics of drugs play a crucial role in formulating stable and bioavailable drug products. The solubility of a drug, governed by the varying pH along the GI tract and its dissociation constant (pKa), is pivotal in determining its ionization state and absorption rate. Notably, weak acids and bases remain unionized and are absorbed more rapidly.
Enhanced drug absorption can be achieved by reducing particle sizes and increasing surface areas, thereby facilitating...

Structure of Lipids03:38

Structure of Lipids

101.4K
Lipids include a diverse group of compounds that are largely nonpolar in nature. This is because they are hydrocarbons that include mostly nonpolar carbon-carbon or carbon-hydrogen bonds. Non-polar molecules are hydrophobic (“water fearing”), or insoluble in water. Lipids perform many different functions in a cell. Cells store energy for long-term use in the form of fats. Lipids also provide insulation from the environment for plants and animals. For example, they help keep aquatic...

Factors Affecting Dissolution: Drug Permeability, Stability and Stereochemistry

665
Orally administered drugs primarily enter the systemic circulation via passive diffusion through the intestinal membranes. The drug's absorption is influenced by drug stability in the gastrointestinal GI tract, membrane permeability, the surface area available for absorption, luminal drug concentration, and residence time in the lumen. Drug permeability can be enhanced by adjusting the lipophilicity, polarity, or molecular size of the drug, promoting its passive transport across intestinal...

Lipid-Lowering Drugs: Statins and Miscellaneous Agents

1.8K
Hyperlipidemia, a medical condition often referred to as high cholesterol, is characterized by abnormally elevated levels of lipids in the bloodstream. When present in excess, these lipids, specifically cholesterol and triglycerides, can lead to serious health complications, often involving cardiovascular diseases. Illnesses like atherosclerosis, heart attacks, and pancreatitis have all been linked to untreated hyperlipidemia. This means controlling and regulating cholesterol and triglyceride...

Area of Science:

  • Pharmaceutical Technology
  • Nutraceutical Science
  • Materials Science

Background:

  • Omega-3 fatty acids are widely consumed as health supplements.
  • Various delivery forms exist, but omega-3 tablets remain an underexplored niche.
  • Developing stable and effective omega-3 tablet formulations is challenging.

Purpose of the Study:

  • To investigate the feasibility of producing omega-3 tablets via direct compression.
  • To evaluate the role of beta-cyclodextrin as an encapsulating agent for omega-3 oils.
  • To characterize the physical properties and crystalline structure of the developed omega-3 tablets.

Main Methods:

  • Preparation of omega-3 oil powders (triglycerides or ethyl esters) with beta-cyclodextrin.
  • Direct compression of prepared powders into tablets.
  • Analysis of powder and tablet physical properties.
  • X-ray diffractometry and proton nuclear magnetic resonance ((1)H NMR) spectroscopy for structural analysis.

Main Results:

  • Tablets with up to 35% (w/w) triglyceride oil and 30% (w/w) ethyl ester oil were successfully produced with excellent quality.
  • X-ray diffraction indicated the formation of new crystalline phases.
  • (1)H NMR data suggested inclusion complex formation between beta-cyclodextrin and omega-3 ethyl esters.
  • Commercially available omega-3 powders failed direct compression.

Conclusions:

  • Direct compression is a viable method for producing high-quality omega-3 tablets using beta-cyclodextrin encapsulation.
  • Beta-cyclodextrin facilitates the formation of stable omega-3 formulations with unique crystalline structures.
  • This approach offers a promising new administration route for omega-3 nutraceuticals.
