Ribosome Profiling

4.3K
Ribosome profiling or ribo-sequencing is a deep sequencing technique that produces a snapshot of active translation in a cell. It selectively sequences the mRNAs protected by ribosomes to get an insight into a cell’s translation landscape at any given point in time.
Applications of ribosome profiling
Ribosome profiling has many applications, including in vivo monitoring of translation inside a particular organ or tissue type and quantifying new protein synthesis levels.

Genomic DNA in Eukaryotes

54.0K
Eukaryotes have large genomes compared to prokaryotes. To fit their genomes into a cell, eukaryotic DNA is packaged extraordinarily tightly inside the nucleus. To achieve this, DNA is tightly wound around proteins called histones, which are packaged into nucleosomes that are joined by linker DNA and coil into chromatin fibers. Additional fibrous proteins further compact the chromatin, which is recognizable as chromosomes during certain phases of cell division.

Area of Science:

  • Genomics
  • Bioinformatics
  • Molecular Biology

Background:

  • The human genome is organized into isochores, DNA sequences with homogeneous base composition.
  • Mammalian genome compartmentalization has been studied for decades, yet automatic isochore segmentation remains challenging.

Purpose of the Study:

  • To critically evaluate existing genome segmentation methods.
  • To introduce isoSegmenter, a novel, fast, and fully automatic approach for isochore segmentation.

Main Methods:

  • Developed isoSegmenter, an algorithm for genome segmentation into isochores.
  • Utilized experimentally defined parameters: compositional boundaries of isochore families and an optimal 100 kb window size.

Main Results:

  • isoSegmenter provides fast and automatic genome segmentation into isochores.
  • The method improves upon existing genome segmentation techniques.

Conclusions:

  • isoSegmenter is a significant advancement for analyzing long-range genomic features in sequenced and assembled genomes.
  • The tool is publicly available for researchers to study genome organization.
