    Summary

    This study investigated ferritin in acute myeloid leukaemia (AML) cells. Results indicate no specific ferritin pattern for AML diagnosis, but abnormal subunits may be relevant for future research.

    Background:

    • Ferritin, an iron-storage protein, exhibits isoferritins varying by tissue and disease state.
    • Previous research suggested specific

    • isoferritins in certain cancers.

    Purpose of the Study:

    • To analyze the isoferritin profiles of ferritin from acute myeloid leukaemia (AML) cells.
    • To determine if specific isoferritin patterns can aid in AML diagnosis or treatment monitoring.

    Main Methods:

    • Partial purification of ferritin from leucocyte extracts of AML patients and normal subjects.
    • Anion-exchange chromatography and isoelectric focusing were employed for isoferritin analysis.

    Main Results:

    • Leucocyte ferritin iron content was generally low.
    • Most AML ferritin preparations contained isoferritins similar to normal tissues or serum.
    • Only some preparations showed acidic isoferritins also found in normal heart and kidney.
    • One patient's ferritin displayed unique, very acidic isoferritins.

    Conclusions:

    • No specific isoelectric focusing pattern for leukaemic cells was identified.
    • Assays for acidic isoferritins are unlikely to be useful for diagnosing or monitoring AML.
    • The presence of highly acidic ferritin in one case suggests potential for abnormal ferritin subunits in AML.
