High-voltage paper electrophoretic assay for guanyl cyclase.

H Mukerjee

    Journal of Chromatography

    PubMed
    Summary

This study presents a fast and sensitive method using high-voltage paper electrophoresis to separate cyclic guanosine monophosphate (cGMP) from other guanine nucleotides. The technique demonstrates high recovery rates for labeled cGMP in guanylyl cyclase assays with minimal background noise.

    Area of Science:

    • Biochemistry
    • Analytical Chemistry

    Background:

    • Guanine nucleotides, including cyclic guanosine monophosphate (cGMP), play crucial roles in cellular signaling pathways.
    • Accurate separation and quantification of cGMP are essential for studying its biological functions and related enzyme activities.

    Purpose of the Study:

    • To develop a simple, sensitive, and rapid method for separating cyclic guanosine monophosphate (cGMP) from guanosine monophosphate (GMP), guanosine diphosphate (GDP), and guanosine triphosphate (GTP).

    Main Methods:

    • High-voltage paper electrophoresis was employed for the unidirectional separation of guanine nucleotides.
    • The method was validated using labeled cGMP in guanylyl cyclase assays.

    Main Results:

    • The described technique effectively separates cGMP from GMP, GDP, and GTP.
    • Recovery of labeled cGMP in guanylyl cyclase assays was high, ranging from 85% to 90%.
    • Blank values, representing non-enzymatic background, were found to be negligible.

    Conclusions:

    • High-voltage paper electrophoresis provides a robust and efficient method for cGMP analysis.
    • This technique is suitable for accurate determination of guanylyl cyclase activity.
