BCL11A expression in acute myeloid leukemia.

High BCL11A expression in acute myeloid leukemia (AML) correlates with specific clinical and laboratory features. While not significantly impacting remission or survival rates, BCL11A may play a role in AML biology.

Purpose of the Study:
- To investigate the relationship between BCL11A expression levels at diagnosis and clinical/laboratory characteristics in newly diagnosed AML patients.

- BCL11A mRNA levels were quantified using reverse transcription and real-time PCR in bone marrow and blood mononuclear cells from 292 newly diagnosed AML patients.
- BCL11A levels were compared to those in normal bone marrow cells.
- Correlations with clinical, laboratory, and cytogenetic variables, as well as treatment outcomes, were analyzed.

- Higher BCL11A transcript levels at diagnosis were associated with increased white blood cell counts, a higher proportion of bone marrow myeloblasts, FAB M0 subtype, intermediate cytogenetic risk, and the presence of DNMT3A(R882) and FLT3-ITD mutations.
- Conversely, higher BCL11A levels were linked to a lower proportion of FAB M3 subtype and less frequent complex karyotypes.
- In patients receiving chemotherapy, complete remission rates were 54% (lower BCL11A) vs. 65% (higher BCL11A), and 3-year survival was 33% vs. 15%, respectively, with non-significant differences.
Conclusions:
- BCL11A transcript levels at AML diagnosis show significant associations with various clinical and laboratory parameters.
- While not reaching statistical significance, trends suggest potential non-significant associations with complete remission rates and survival.
- These findings indicate a potential role for BCL11A expression in the biological mechanisms underlying AML.
