Summary

Leptin regulates reproduction by signaling through intermediate neurons. Researchers found that leptin-responsive, GABAergic neurons in the arcuate nucleus project to GnRH neurons, suggesting their role in this signaling pathway.

Background:

  • Leptin, an adipocyte hormone, is crucial for reproductive control.
  • Gonadotropin-releasing hormone (GnRH) neurons, key regulators of the reproductive axis, lack leptin receptors.
  • Leptin signaling to GnRH neurons necessitates intermediary leptin-responsive neurons.

Purpose of the Study:

  • To identify leptin-responsive neuronal populations projecting to the rostral preoptic area (rPOA), the location of GnRH cell bodies.
  • To investigate the role of GABAergic neurons within these leptin-responsive populations in mediating leptin-to-GnRH signaling.

Main Methods:

  • Injected fluorescent retrograde tracer beads into the rPOA of Lepr-eGFP reporter mice to label projecting neurons.
  • Assessed tracer uptake in Lepr-eGFP positive neurons across hypothalamic nuclei (ARC, DMN, PMV).
  • Repeated experiments in Vgat-tdTomato reporter mice to identify GABAergic neurons and their projection patterns.

Main Results:

  • Retrograde tracer uptake was highest in leptin receptor-expressing neurons within the medial arcuate nucleus (ARC).
  • A significant proportion (10-45%) of tracer-labeled neurons in the ARC were GABAergic.
  • GABAergic neuron projection to the rPOA was minimal in the dorsomedial nucleus (DMN) and ventral premammillary nucleus (PMV).

Conclusions:

  • Leptin-responsive and GABAergic neurons in the ARC project to the vicinity of GnRH neurons.
  • These ARC-derived LEPR-expressing GABAergic neurons are potential mediators of leptin's reproductive signaling to GnRH neurons.
