Background And Aims:
Dysregulation of microRNAs (miRNAs) is associated with a variety of diseases, including Crohn's disease (CD), but the essential biological functions and crucial targets of miRNAs remain largely unknown. The present study investigated the aberrant colonic mucosal miRNAs in active CD patients.
Methods:
miRNA levels were assayed in inflamed colon of active CD patients by quantitative real-time polymerase chain reaction. The influence of differential expressed miR-124 on its putative target, the aryl hydrocarbon receptor (AHR), was investigated in CD patients, intestinal epithelial cells (IECs) and 2,4,6-trinitrobenzene sulphonic acid (TNBS)-induced colitis mice. The role of miR-124 was further studied in experimental colitis mice by intracolonic administration of miR-124 inhibitors or precursors.
Results:
We found an inverse correlation between miR-124 and AHR protein levels in colon tissues and IECs of active CD patients. Further results demonstrated that miR-124 suppressed AHR expression by directly targeting the AHR 3'-untranslated region (3'-UTR) in Caco-2 cells and HT-29 cells. MiR-124 mediated the inflammatory response in lipopolysaccharide-stimulated cells through retroregulation of AHR in vitro. Downregulation or upregulation of miR-124 in TNBS-induced colitic colon alleviated or aggravated experimental colitis, respectively.
Conclusions:
These findings suggest that miR-124 induces intestinal inflammation by inhibiting AHR to modulate pro-inflammatory cytokine production and thereby promotes the pathogenesis of CD.
Insights
MicroRNA-124 (miR-124) promotes intestinal inflammation in Crohn's disease (CD) by inhibiting the aryl hydrocarbon receptor (AHR). This study reveals miR-124 as a key player in CD pathogenesis.
Area of Science:
- Gastroenterology
- Molecular Biology
- Immunology
Background:
- MicroRNA (miRNA) dysregulation is implicated in Crohn's disease (CD) pathogenesis.
- The specific roles and targets of miRNAs in CD remain largely uncharacterized.
- This study focuses on aberrant colonic mucosal miRNAs in active CD patients.
Purpose of the Study:
- To investigate the role of microRNA-124 (miR-124) in the colonic mucosa of active Crohn's disease (CD) patients.
- To determine the relationship between miR-124 and its target, aryl hydrocarbon receptor (AHR).
- To elucidate the functional impact of miR-124 on intestinal inflammation and CD pathogenesis.
Main Methods:
- Quantitative real-time polymerase chain reaction (PCR) to assay miRNA levels in inflamed colon tissue.
- Investigation of miR-124 and AHR interaction in intestinal epithelial cells (IECs) and a mouse model of colitis.
- In vivo studies using miR-124 inhibitors and precursors in experimental colitis models.
Main Results:
- An inverse correlation was observed between miR-124 and AHR protein levels in active CD patients.
- miR-124 directly targets the AHR 3'-untranslated region (3'-UTR), suppressing AHR expression in intestinal cells.
- Modulation of miR-124 levels in experimental colitis significantly altered disease severity, indicating its regulatory role in inflammation.
Conclusions:
- miR-124 inhibition of AHR contributes to intestinal inflammation by affecting pro-inflammatory cytokine production.
- This miR-124/AHR axis plays a crucial role in promoting the pathogenesis of Crohn's disease.
- miR-124 represents a potential therapeutic target for managing CD-associated intestinal inflammation.

Inflammatory Bowel Disease II: Crohn's Disease
Inflammatory bowel disease, commonly known as IBD, refers to a collection of disorders that lead to persistent inflammation of the gastrointestinal tract. The two types of IBD are ulcerative colitis, which impacts the colon, and Crohn's disease, which can involve any part of the gastrointestinal segment.
Crohn's disease
Crohn's disease is a chronic, systemic inflammatory bowel disease (IBD) that predominantly affects the gastrointestinal tract. It is marked by...
