Area of Science:

  • Analytical Chemistry
  • Food Science
  • Biochemistry

Background:

  • A novel assay quantifies singlet oxygen absorption capacity (SOAC) in natural antioxidants and food extracts.
  • Traditional SOAC measurements utilize UV-Vis spectrophotometry under specific solution conditions (ethanol-chloroform-D2O at 35°C).

Purpose of the Study:

  • To evaluate the applicability of a microplate reader for measuring SOAC values.
  • To compare SOAC measurements obtained using a microplate reader with those from a UV-Vis spectrophotometer.

Main Methods:

  • SOAC values were determined for eight carotenoids and three vegetable extracts (tomato, carrot, red paprika).
  • Measurements were conducted using a microplate reader with a 24-well glass microplate.
  • A specific solvent mixture (ethanol-chloroform-D2O, 50:50:1 v/v/v) was employed, necessitating the use of glass microplates to prevent dissolution.

Main Results:

  • SOAC values obtained via microplate reader showed strong agreement with those measured by UV-Vis spectrophotometry.
  • The microplate reader proved to be a reliable instrument for SOAC quantification.

Conclusions:

  • The microplate reader is a suitable and versatile instrument for measuring SOAC in various antioxidants and food extracts.
  • This finding supports the use of microplate readers for routine and reliable SOAC analysis in research and quality control settings.
