The new compound NIOCH-14 shows potent antiviral activity against orthopoxviruses, comparable to ST-246. This promising antiviral agent demonstrates efficacy in vivo, suggesting potential for a new smallpox drug.
Area of Science:
- Virology
- Medicinal Chemistry
- Pharmacology
Background:
- Orthopoxviruses pose significant public health threats, necessitating the development of novel antiviral therapies.
- Existing treatments have limitations, driving the search for effective alternatives against highly pathogenic strains.
Purpose of the Study:
- To evaluate the antiviral efficacy of a novel compound, NIOCH-14, against orthopoxviruses.
- To compare the activity of NIOCH-14 with the established antiviral ST-246 in vitro and in vivo.
- To assess the therapeutic window and safety profile of NIOCH-14.
Main Methods:
- In vitro and in vivo antiviral assays using various orthopoxviruses (ectromelia, monkeypox, variola).
- Oral administration of NIOCH-14 and ST-246 to infected animal models (ICR mice, SCID mice, marmots).
- Quantification of viral titers, assessment of pathological changes, and survival rates.
Main Results:
- NIOCH-14 demonstrated comparable 50% effective doses to ST-246 against ectromelia virus.
- Both compounds significantly reduced viral loads and pathological lung changes in infected mice and marmots.
- A broad therapeutic window was observed for NIOCH-14, extending from before infection to days post-infection.
Conclusions:
- NIOCH-14 exhibits significant antiviral activity against a range of orthopoxviruses, including highly pathogenic strains.
- The compound shows promise as a potential therapeutic agent for diseases caused by orthopoxviruses.
- Further research into NIOCH-14 is warranted for the development of a new smallpox drug.
