This study developed a bioactive hydrogel using insulin-like growth factor-1 (IGF-1) to improve stem cell therapy for acute kidney injury (AKI). The engineered hydrogel enhanced adipose-derived mesenchymal stem cell (ADSC) survival and promoted kidney repair in AKI models.
Area of Science:
- Biomaterials Science
- Regenerative Medicine
- Nephrology
Background:
- Stem cell therapy for acute kidney injury (AKI) faces challenges with low cell retention and engraftment.
- Engineered microenvironments, including hydrogels, can mimic stem cell niches to improve therapeutic outcomes.
- Insulin-like growth factor-1 (IGF-1) plays a role in cell survival and tissue repair.
Purpose of the Study:
- To synthesize a bioactive hydrogel by immobilizing the C domain peptide of IGF-1 (IGF-1C) on chitosan.
- To investigate if the chitosan-IGF-1C hydrogel enhances adipose-derived mesenchymal stem cell (ADSC) survival and function in an AKI model.
- To evaluate the therapeutic potential of this engineered hydrogel for AKI treatment.
Main Methods:
- Synthesis of a chitosan-IGF-1C bioactive hydrogel.
- In vitro assessment of ADSC viability and paracrine effects within the hydrogel.
- In vivo transplantation of ADSCs with the hydrogel into an ischemic AKI model.
- Evaluation of renal function, cell survival, angiogenesis, and fibrosis using bioluminescence imaging and histological analysis.
Main Results:
- The chitosan-IGF-1C hydrogel significantly increased ADSC viability in vitro compared to controls.
- In vivo studies showed ameliorated renal function in AKI models treated with chitosan-IGF-1C hydrogel and ADSCs.
- Enhanced stem cell survival, increased angiogenesis, and reduced renal fibrosis were observed in the treated kidneys.
Conclusions:
- Immobilizing IGF-1C on a chitosan hydrogel creates a supportive microenvironment for ADSCs.
- This engineered hydrogel promotes stem cell survival and therapeutic efficacy in AKI.
- The chitosan-IGF-1C hydrogel represents a promising strategy for stem cell-based AKI therapy.
