Estrogen receptor alpha (ERα) is a newly discovered regulator of renin gene expression. ERα directly binds to the renin enhancer, maintaining renin levels in cells.
Area of Science:
- Endocrinology
- Molecular Biology
- Genetics
Background:
- Renin-angiotensin-aldosterone system (RAAS) regulates blood pressure.
- Renin's enzymatic activity is crucial for angiotensin II production.
- Mechanisms controlling renin gene expression are not fully understood.
Purpose of the Study:
- To elucidate the molecular mechanisms governing renin gene expression.
- To identify novel regulators of renin expression.
Main Methods:
- Bioinformatic analysis of microarray data.
- Small interfering RNA (siRNA) transfection in renin-expressing cells (As4.1).
- Quantitative reverse transcription PCR (qRT-PCR) to assess gene expression.
- Chromatin immunoprecipitation (ChIP) assay to determine receptor binding.
Main Results:
- Estrogen receptor alpha (ERα) and hepatic nuclear factor 4α were identified as potential regulators.
- ERα is essential for maintaining renin expression, confirmed by siRNA knockdown.
- ERα regulates renin independently of known negative regulators (Nr2f2, vitamin D receptor).
- ERα directly binds to a hormone response element in the renin enhancer region.
Conclusions:
- Estrogen receptor alpha (ERα) is a novel, direct regulator of renin gene expression.
- ERα plays a critical role in maintaining renin expression via direct enhancer binding.
