  • Accurate strain typing tools are essential for the industrial application of C. maltaromaticum.

Purpose of the Study:

  • To develop and validate a novel, high-resolution typing method for C. maltaromaticum.
  • To assess the discriminatory power of the new method compared to existing techniques.

Main Methods:

  • A Repeat-based Sequence Typing (RST) method was developed, analyzing three Variable-Number Tandem-Repeat (VNTR) loci.
  • PCR amplification of VNTR loci followed by sequence analysis was performed on 24 C. maltaromaticum strains.

Main Results:

  • Sequence analysis identified 11, 11, and 12 alleles for VNTR-A, VNTR-B, and VNTR-C, respectively.
  • The RST method defined 15 distinct genotypes with a high discriminatory index of 0.94.
  • Comparison with Multilocus Sequence Typing (MLST) indicated complementary typing capabilities.

Conclusions:

  • The novel RST method provides accurate and high-resolution strain typing for C. maltaromaticum.
  • This method enhances the industrial utilization of C. maltaromaticum in food applications, particularly in the dairy sector.
