This study developed a novel gastro retentive floating drug delivery system for improved oral drug bioavailability. The optimized formulation demonstrated enhanced bioadhesion and sustained drug release over 24 hours.
Area of Science:
- Pharmaceutical Sciences
- Drug Delivery Systems
Background:
- Gastro retentive dosage forms enhance bioavailability for drugs with narrow absorption windows.
- Floating bioadhesive systems prolong gastric residence time.
Purpose of the Study:
- To develop a floating bioadhesive drug delivery system for sustained drug release.
- To prolong the gastric residence time of medications using atenolol as a model drug.
Main Methods:
- Tablets prepared via direct compression using Carbopol 934P and HPMC polymers.
- Evaluation of polymeric blends for flow properties.
- Assessment of tablets for physical characteristics, bioadhesion, buoyancy, swelling, and in vitro drug release.
Main Results:
- Optimized formulation (F11) achieved 92.3% drug release in 24 hours.
- Increased Carbopol 934P concentration enhanced bioadhesive strength and swelling index, leading to slower release.
- Non-fickian anomalous transport mechanism observed (n values 0.631-0.719).
Conclusions:
- Successfully developed a once-daily gastro retentive floating drug delivery system.
- The system exhibited improved floating, swelling, and bioadhesive properties.
- Enhanced bioavailability is expected due to prolonged drug release and gastric residence.
