Polytene Chromosomes02:04

Polytene Chromosomes

11.3K
Polytene chromosomes are giant interphase chromosomes with several DNA strands placed side by side. They were discovered in the year 1881 by Balbiani in salivary glands, intestine, muscles, malpighian tubules, and hypoderm of larvae Chironomus plumosus. Hence, these are also called "Salivary gland chromosomes." These are found in insects of the order Diptera and Collembola; in certain organs of mammals; and synergids, antipodes of flowering plants. Polytene chromosomes are also...

Lampbrush Chromosomes

8.8K
In 1882, Flemming observed lampbrush chromosomes (LBC) in salamander eggs. Later in 1892, Rückert observed LBCs in shark egg cells and coined the term "lampbrush chromosomes" because they looked like brushes used to clean kerosene lamps.
LBCs are made up of two pairs of conjugating homologous chromatids. Each chromatid consists of alternatively positioned regions of condensed-inactive chromatin and loosely placed-active side loops, which can be contracted and extended. The loops...

Area of Science:

  • Molecular Biology
  • Genetics
  • Biochemistry

Background:

  • Astrephomene gubernaculilera is a species with a distinct 7-chromosome structure.
  • Understanding the genetic makeup of novel organisms is crucial for evolutionary and biological studies.

Purpose of the Study:

  • To isolate, partially purify, and characterize the DNA from Astrephomene gubernaculilera.
  • To determine the guanine-cytosine (G-C) content of the organism's DNA.

Main Methods:

  • Enzymatic removal of sheath material from individual cells.
  • Cell lysis using hot sodium lauryl sulfate.
  • DNA isolation and purification via a modified Marmur procedure.
  • Melting experiments and buoyant-density measurements for G-C content analysis.

Main Results:

  • Successfully isolated and purified DNA from Astrephomene gubernaculilera.
  • Indicated a native DNA guanine-cytosine (G-C) content of 61-62%.
  • Identified a stoichiometric presence of a pentose phosphorus-containing compound.

Conclusions:

  • The study provides the first characterization of DNA from Astrephomene gubernaculilera.
  • The high G-C content suggests specific evolutionary or functional adaptations.
  • Further research can build upon these foundational genetic insights.
