Background:

  • Uninephrectomy (kidney removal) and high-protein diets can induce significant physiological changes in the remaining kidney.
  • Sex-based differences in susceptibility to kidney damage are increasingly recognized, but underlying mechanisms require further elucidation.

Purpose of the Study:

  • To investigate sex-based differences in glomerular hemodynamics following uninephrectomy.
  • To assess the long-term impact of repeated pregnancies on renal function in uninephrectomized female rats.
  • To evaluate the utility of an acute glycine challenge test in assessing glomerular damage.

Main Methods:

  • Micropuncture measurements were used to determine glomerular capillary blood pressure (BP) in male and female rats at 9-10 months post-uninephrectomy.
  • A separate cohort of female rats underwent chronic uninephrectomy followed by multiple pregnancies and lactations.
  • An acute intravenous glycine challenge was administered to assess renal response.

Main Results:

  • Male rats showed significantly higher glomerular capillary BP compared to virgin female rats 9-10 months after uninephrectomy.
  • Despite higher BP, significant differences in renal function or glomerulosclerosis were minimal between sexes at this time point.
  • Multiple pregnancies and lactations in uninephrectomized females did not result in severe long-term deleterious effects on the glomeruli.
  • Glycine challenge responses were variable and did not consistently correlate with the extent of glomerular damage.

Conclusions:

  • Elevated glomerular capillary BP in male rats post-uninephrectomy may predispose them to greater renal damage.
  • Pregnancy appears to be relatively safe for the glomeruli in female rats with pre-existing renal compromise.
  • The acute glycine challenge test may have limited value in assessing underlying glomerular disease in this model.
