Background:

  • The intergeniculate leaflet (IGL) contains neuropeptide Y-immunoreactive (NPY-IR) neurons projecting to the suprachiasmatic nucleus (SCN).
  • The pregeniculate nucleus is considered the primate homologue of the rodent IGL.

Purpose of the Study:

  • To investigate the organization of NPY-IR neurons and their projections in the primate SCN.
  • To compare the human SCN with that of other mammals, including macaques.

  • Immunohistochemistry to identify NPY-IR neurons and axons.
  • Comparative analysis of brain tissue from rodents, macaque monkeys, and humans.

  • Macaque monkeys exhibit NPY-IR neurons in the pregeniculate nucleus and a dense NPY-IR axon plexus in the SCN, similar to rodents.
  • Humans possess NPY-IR neurons within the SCN itself, unlike macaques and rodents.

Conclusions:

  • The human SCN shows a distinct organization compared to other mammals.
  • Local NPY+ neurons within the human SCN may serve as the functional homologue of the mammalian geniculohypothalamic tract.
