Avanafil, a second-generation phosphodiesterase type 5 inhibitor (PDE5I), offers effective treatment for erectile dysfunction (ED) with rapid onset. Its high selectivity leads to comparable efficacy and potentially fewer side effects than older PDE5Is.
Area of Science:
- Pharmacology
- Urology
Background:
- Phosphodiesterase type 5 inhibitors (PDE5Is) are first-line treatments for erectile dysfunction (ED).
- A significant portion of ED patients experience treatment dissatisfaction or lack of response to existing PDE5Is.
- Avanafil is a recently developed second-generation PDE5I for ED treatment.
Purpose of the Study:
- To review the clinical development and efficacy of avanafil for erectile dysfunction.
- To analyze the pharmacokinetics and pharmacodynamics of avanafil.
- To compare avanafil's safety and efficacy profile with existing PDE5Is.
Main Methods:
- Systematic literature search of English-language studies up to May 2016 in the Medline database.
- Inclusion of pivotal clinical development studies and placebo-controlled randomized clinical trials (RCTs) of avanafil in ED patients.
- Analysis of pharmacokinetic and pharmacodynamic studies related to avanafil.
Main Results:
- Avanafil demonstrates efficacy comparable to existing PDE5Is in both treatment-naïve and previously treated ED patients.
- Avanafil is effective approximately 15 minutes after dosing, uniquely approved for use 15-30 minutes before sexual activity.
- High selectivity for the PDE5 isoenzyme results in a lower incidence of drug-related side effects compared to other PDE5Is.
Conclusions:
- Avanafil is a potent and highly selective PDE5I for ED treatment.
- Its rapid onset of action and favorable side effect profile offer a valuable alternative for ED management.
- Avanafil provides comparable efficacy to existing PDE5Is with potential for improved patient satisfaction.
