A new method uses mass-barcoded gold nanoparticles (AuNPs) and immunomagnetic separation for sensitive, component-resolved allergy diagnosis. This approach allows simultaneous identification of multiple specific IgE antibodies (Abs) for improved diagnostic efficiency.
Area of Science:
- Biotechnology
- Analytical Chemistry
- Immunology
Background:
- Allergy diagnosis often requires component-resolved analysis to identify specific IgE antibodies (Abs).
- Existing methods may lack sensitivity or the ability to perform multiplex testing efficiently.
- There is a need for highly sensitive diagnostic tools, especially when sample volumes are limited.
Purpose of the Study:
- To develop a highly sensitive and multiplex method for component-resolved allergy diagnosis.
- To utilize mass-barcoded gold nanoparticles (AuNPs) and immunomagnetic separation for IgE Ab detection.
- To achieve a low limit of detection for specific IgE Abs in patient serum.
Main Methods:
- Preparation of AuNPs coated with allergenic proteins and mass barcodes (polyethylene glycol).
- Immunomagnetic separation using magnetic beads (MBs) functionalized with anti-human IgE antibodies (Abs).
- Detection of specific IgE Abs via mass spectrometry analysis of mass barcodes in a sandwich complex.
Main Results:
- Simultaneous identification of multiple specific IgE Abs was achieved.
- The method demonstrated high sensitivity with a limit of detection down to picograms per milliliter.
- The assay utilizes signal amplification from mass barcodes for enhanced detection.
Conclusions:
- The developed method offers a sensitive and efficient approach for component-resolved allergy diagnosis.
- This technique is particularly valuable for multiplex testing with limited sample amounts.
- The use of mass-barcoded AuNPs and immunomagnetic separation shows significant potential in allergy diagnostics.
