Summary

Thyroid cell cultures retain characteristics for 21 days. After this, fibroblast-like cells emerge, losing key thyroid markers. Optimal in vitro thyroid research conditions require careful consideration of culture duration.

Background:

  • Thyroid cell models are crucial for in vitro research.
  • Permanent cell lines often originate from tumors, necessitating comparison with normal thyroid cells.
  • Primary normal thyroid cultures have limitations, including slow growth, limited lifespan, and loss of thyroid characteristics.

Purpose of the Study:

  • To evaluate coding and noncoding thyroid markers in different culture systems.
  • To determine optimal conditions for in vitro thyroid experimentation using primary cells.

Main Methods:

  • Primary human thyroid cells were cultured in two media (6H and h7H).
  • Cell morphology and gene expression (9 key thyroid markers) were monitored via RT-PCR at days 8, 21, and 43.
  • A pilot study used a 3D hanging-drop culture system.

Main Results:

  • Primary thyroid cells grew and retained morphology for the first two weeks.
  • Most thyroid gene markers were present by day 21, but declined by day 43.
  • lncRNA transcripts PTCSC2 and PTCSC3 were the earliest markers to be lost. The 3D system retained markers longer by day 21.

  • Thyroid cell cultures maintain characteristics up to 21 days.
  • Dedifferentiation occurs with the emergence of fibroblast-like cells after 21 days.
