Cell Lines01:16

Cell Lines

11.1K
A cell line is a population of cells grown in vitro that can be subcultured over several generations. Normal cells cease to divide after a certain number of cell divisions, a process known as replicative senescence. This number, called the Hayflick limit, was conceptualized by Leonard Hayflick in 1961 when he observed that fetal cells grown in culture could only divide 40-60 times. This limit is due to the shortening of the telomeres during each round of cell division, preventing cell division...

Area of Science:

  • Genomics
  • Biotechnology
  • Cell Biology

Background:

  • Cell line authentication is critical for reproducible research.
  • Short tandem repeat (STR) profiling is the only established standard (ASN-0002) for cell line authentication.
  • Various genomic technologies exist for cell line analysis.

Purpose of the Study:

  • To highlight the importance of the ASN-0002 standard for cell line authentication.
  • To discuss the critical role of standards in advancing cell line-based research.
  • To explore alternative and complementary methods for comprehensive cell line quality assessment.

Main Methods:

  • Review of existing genomic technologies for cell line authentication.
  • Analysis of the ASN-0002 standard for STR profiling.
  • Discussion of a holistic approach combining multiple authentication methods.

Main Results:

  • Short tandem repeat (STR) profiling is the sole method with a comprehensive standard (ASN-0002).
  • Standards are essential for establishing consensus technical criteria and practices.
  • A combination of methods can provide a more complete assessment of cell line quality.

Conclusions:

  • The ASN-0002 standard significantly advances cell line authentication through STR profiling.
  • Standardization is key to enabling progress in cell line research.
  • A multi-method approach offers a holistic strategy for ensuring cell line quality and reliability.
