Coefficient of Correlation01:12

Coefficient of Correlation

9.0K
The correlation coefficient, r, developed by Karl Pearson in the early 1900s, is numerical and provides a measure of strength and direction of the linear association between the independent variable x and the dependent variable y.
If you suspect a linear relationship between x and y, then r can measure how strong the linear relationship is.
What the VALUE of r tells us:
The value of r is always between –1 and +1: –1 ≤ r ≤ 1.
The size of the correlation r indicates the...

Mutation, Gene Flow, and Genetic Drift01:09

Mutation, Gene Flow, and Genetic Drift

65.6K
In a population that is not at Hardy-Weinberg equilibrium, the frequency of alleles changes over time. Therefore, any deviations from the five conditions of Hardy-Weinberg equilibrium can alter the genetic variation of a given population. Conditions that change the genetic variability of a population include mutations, natural selection, non-random mating, gene flow, and genetic drift (small population size).

Genetic Variation01:25

Genetic Variation

1.5K
Genetic variation is the diversity in DNA sequences found among individuals of the same species. This diversity is crucial for a species' survival because it helps organisms adapt to environmental changes. Genetic variation begins with fertilization, where an egg and sperm cell merge. Each of these cells carries 23 chromosomes, up to 46 in the fertilized egg. Chromosomes are long DNA strands that contain genes, the basic units of heredity.
Genes exist in different versions called alleles,...

Test for Homogeneity01:23

Test for Homogeneity

2.5K
The goodness–of–fit test can be used to decide whether a population fits a given distribution, but it will not suffice to decide whether two populations follow the same unknown distribution. A different test, called the test for homogeneity, can be used to conclude whether two populations have the same distribution. To calculate the test statistic for a test for homogeneity, follow the same procedure as with the test of independence. The hypotheses for the test for homogeneity can...

Correlation of Experimental Data01:23

Correlation of Experimental Data

511
Dimensional analysis simplifies complex physical problems and guides experimental investigations, but it does not provide complete solutions. It identifies the dimensionless groups that influence a phenomenon, but experimental data is needed to establish the specific relationships and validate theoretical predictions.
For example, a spherical particle moving through a viscous fluid experiences drag. Dimensional analysis shows that the drag force depends on the particle's diameter, velocity,...

Genetics of Speciation02:16

Genetics of Speciation

23.0K
Speciation is the evolutionary process resulting in the formation of new, distinct species—groups of reproductively isolated populations.

Summary

Genetic correlation between populations for complex traits is often less than 1. Our new method uses genome-wide association studies summary data to estimate this transethnic genetic correlation, revealing variations in genetic architecture across diverse groups.

Area of Science:

  • Population Genetics
  • Medical Genetics
  • Statistical Genetics

Background:

  • Genetic association studies across multiple populations offer insights into the variation of complex trait genetic architecture.
  • Understanding population-specific genetic architecture is crucial for both medical and population genetics applications.

Purpose of the Study:

  • To develop a novel method for estimating transethnic genetic correlation.
  • To assess the correlation of causal-variant effect sizes at single nucleotide polymorphisms (SNPs) common across populations.

Main Methods:

  • Developed a method utilizing the full spectrum of SNP associations from genome-wide association studies (GWAS) summary-level data.
  • The approach avoids genotype-level data, mitigating computational costs and privacy concerns.
  • The method is scalable to large datasets (hundreds of thousands of individuals, millions of SNPs).

Main Results:

  • Applied the method to gene expression, rheumatoid arthritis, and type 2 diabetes data.
  • Consistently found that transethnic genetic correlation was significantly less than 1 across these diverse traits and populations.
  • Indicates substantial variation in the genetic architecture of complex phenotypes between populations.

Conclusions:

  • The developed method provides an efficient and scalable way to estimate transethnic genetic correlation using GWAS summary data.
  • Significant deviations from a genetic correlation of 1 highlight population-specific genetic influences on complex traits.
  • The method is implemented in a Python package named Popcorn.
