Role of Ephrin-Eph Signalling in Intestinal Stem Cell Renewal01:22

Role of Ephrin-Eph Signalling in Intestinal Stem Cell Renewal

2.8K
Erythropoietin-producing hepatocellular carcinoma receptor (Eph) and its ligand, Eph receptor-interacting protein (Ephrin) were first discovered in the human carcinoma cell line, hence the name. Ephrin-Eph interaction guides cells to reach their appropriate location in adult tissues. They also play an essential role in the immune system by helping in immune cell migration, adhesion, and activation. Based on their structure and function, Eph is divided into two classes — EphA and EphB.

Regulation of Angiogenesis and Blood Supply01:24

Regulation of Angiogenesis and Blood Supply

3.9K
Rapidly dividing tumors, embryos, and wounded tissues require more oxygen than usual, lowering the oxygen concentration in the blood. At low oxygen or hypoxic conditions, an oxygen-sensitive transcription factor called the hypoxia-inducible factor 1 or HIF1 is activated. HIF1 is a dimeric protein of alpha (ɑ) and beta (β) subunits.  Under optimal oxygen conditions, HIF1β is present in the nucleus while HIF1ɑ remains in the cytosol. HIF1ɑ is hydroxylated by prolyl...

Notch Signaling Pathway03:14

Notch Signaling Pathway

6.8K
The Notch signaling pathway is a major intracellular signaling pathway that is highly conserved over a broad spectrum of metazoan species. It stands unique from other intracellular signaling mechanisms in animals because notch protein itself acts as the receptor as well as the primary signaling molecule.
The Notch gene came into the limelight in 1914 after the discovery that its mutation in Drosophila melanogaster leads to a serrated (or "notched") wing margin phenotype. It was not...

Nephrons01:10

Nephrons

8.2K
The kidneys are intricate organs with millions of working units known as nephrons. Each nephron features two major structures: the renal corpuscle, which facilitates blood plasma filtration, and the renal tubule, which handles the glomerular filtrate. Blood supply is directly linked to the nephrons. The renal corpuscle consists of the glomerulus, a capillary network, and the Bowman's capsule, a double-walled epithelial structure that encases the glomerulus. The filtering of blood plasma...

Summary

Bone morphogenic protein-2 (BMP-2) significantly increases nephronectin (Npnt) expression in muscle cells. This finding reveals a new regulatory pathway for Npnt, important for bone and muscle development.

Area of Science:

  • Biochemistry
  • Molecular Biology
  • Cell Biology

Background:

  • Nephronectin (Npnt) is an integrin α8β1 ligand crucial for kidney, liver, bone, and muscle development.
  • Previous research indicated Npnt mRNA expression is modulated by cytokines like TGF-β, TNF-α, and OSM.
  • Overexpression of Npnt has been shown to promote osteoblast differentiation.

Purpose of the Study:

  • To investigate the effect of bone morphogenic protein-2 (BMP-2) on nephronectin (Npnt) expression.
  • To elucidate the signaling pathway involved in BMP-2-mediated Npnt regulation.

Main Methods:

  • Utilized a murine skeletal muscle cell line (C2C12).
  • Analyzed Npnt mRNA expression levels.
  • Investigated the role of the BMP-SMAD signaling pathway.

Main Results:

  • Bone morphogenic protein-2 (BMP-2) strongly upregulated Npnt mRNA expression in C2C12 cells.
  • The BMP-SMAD signaling pathway was identified as the mechanism mediating this upregulation.
  • This establishes a novel regulatory link between BMP-2 and Npnt in muscle cells.

Conclusions:

  • BMP-2 is a potent inducer of nephronectin (Npnt) expression in skeletal muscle cells.
  • The BMP-SMAD pathway mediates BMP-2's effect on Npnt.
  • This finding contributes to understanding Npnt regulation and its role in bone and muscle biology.
