Summary

A 5HT2A receptor antagonist, M100907, significantly reduced repetitive grooming in a mouse model of autism spectrum disorder (ASD). This finding suggests potential for treating restricted interests and repetitive behaviors (RRBs) in individuals with ASD.

Background:

  • Autism spectrum disorder (ASD) is characterized by social-communication deficits and restricted interests and repetitive behaviors (RRBs).
  • Currently, effective treatments for RRBs in ASD are limited.
  • The BTBR mouse model exhibits behaviors relevant to ASD, including elevated grooming, a potential model for stereotyped motor behaviors.

Purpose of the Study:

  • To investigate the efficacy of the 5HT2A receptor antagonist M100907 in reducing repetitive grooming behavior in BTBR mice.
  • To determine if M100907 affects general locomotor activity in BTBR mice.

Main Methods:

  • Administration of M100907 (0.01 or 0.1 mg/kg) or vehicle to BTBR and C57BL6/J (B6) mice.
  • Quantification of repetitive grooming behavior.
  • Assessment of locomotor activity following M100907 administration.

  • M100907 at 0.1 mg/kg significantly attenuated repetitive grooming in BTBR mice compared to vehicle controls.
