Total Internal Reflection Fluorescence Microscopy01:05

Total Internal Reflection Fluorescence Microscopy

13.7K
Total internal reflection fluorescence microscopy or TIRF is an advanced microscopic technique used to visualize fluorophores in samples close to a solid surface with a higher refractive index, such as a glass coverslip. TIRF only allows fluorophores in proximity to the solid surface to be excited. When light from a medium with a lower refractive index (such as air) hits the glass coverslip at a critical angle, the light undergoes total internal reflection stead of passing through the glass.

Glassware Calibration01:11

Glassware Calibration

1.8K
Accurate calibration of glassware, such as volumetric flasks, pipettes, and burettes, is essential to ensure accurate measurements in the analytical laboratory. Calibration helps maintain consistency across measurements and prevents errors arising from inaccurate volumes.
Volumetric flasks: Volumetric flasks are designed to prepare aqueous solutions of precise volumes accurately with a calibration line on the neck. To calibrate a volumetric flask, it is important to fill it with distilled...

Masonry Curtain Walls01:20

Masonry Curtain Walls

1.9K
Masonry curtain walls employ brick or stone veneers supported by the building's structure to form an external cladding system that is both aesthetically appealing and functional. These walls are erected through two principal techniques, first by traditional layering of masonry units and second by using prefabricated panels. Traditional construction relies on steel shelf angles attached to the spandrel beam for support, with high-bond mortars ensuring secure attachment of masonry veneer...

    • Art conservation
    • Material science

    Background:

    • Stained glass windows possess complex artistry revealed under varying light.
    • Virtual relighting offers a powerful tool for appreciating this art form.
    • Analyzing stained glass with controlled illumination is challenging due to size and placement.

    Purpose of the Study:

    • To develop novel methods for virtual relighting of stained glass.
    • To acquire the actual light transport properties of stained glass artifacts.
    • To enable realistic rendering of stained glass under diverse lighting conditions.

    Main Methods:

    • Image-based priors and a material-dependent dictionary for light transport were developed.
    • A laboratory setup studied scattering properties of glass samples.
    • Light transport matrices were recovered using compressive sensing or sparse regularization.

    Main Results:

    • The developed methods successfully recovered heterogeneous light transport matrices.
    • The technique preserves volume impurities in the glass.
    • Rendering results were demonstrated on several stained glass artifacts, including the Rose Window of the Cathedral of Lausanne.

    Conclusions:

    • Novel methods enable accurate virtual relighting of stained glass.
    • The approach effectively captures the complex light transport properties of these artifacts.
    • This facilitates enhanced appreciation and digital preservation of stained glass art.
