Flow Cytometry

17.1K
The development of flow cytometry techniques began in 1934 with initial attempts by Andrew Moldavan, a bacteriologist who counted the cells in a flowing capillary system. Moldavan pumped cells through a capillary tube focused under a microscope for visualization. The invention of photometry allowed the measurement of differentially-stained cells, and Louis Kamentsky developed the first multiparameter flow cytometer in 1965 to identify and count the cancer cells in cervical tissue specimens.

Area of Science:

  • Oncology
  • Cell Biology
  • Biochemistry

Background:

  • Receptor tyrosine kinase like orphan receptor 1 (ROR1) is implicated in cancer progression and embryonic development.
  • ROR1's role in acute lymphoblastic leukemia (ALL) necessitates reliable methods for its assessment.

Purpose of the Study:

  • To establish and validate a flow cytometry protocol for quantifying ROR1 surface expression.
  • To provide a standardized method for ROR1 analysis in ALL cell lines.

Main Methods:

  • Utilizing immortalized acute lymphoblastic leukemia cell lines.
  • Employing flow cytometry for cell surface protein detection.
  • Developing a specific staining and analysis protocol for ROR1.

Main Results:

  • A robust protocol for ROR1 surface expression analysis was successfully developed.
  • The protocol allows for quantitative assessment of ROR1 in ALL cell line models.

Conclusions:

  • The described flow cytometry protocol is effective for analyzing ROR1 expression in ALL.
  • This method can facilitate further research into ROR1's function in leukemia.
