Summary

Elevated miRNA-10b in station 8 lymph nodes may predict early pancreatic cancer recurrence. This finding could aid in assessing disease progression risk for patients undergoing pancreatoduodenectomy.

Background:

  • Lymph node metastasis in pancreatic adenocarcinoma (PAC) impacts survival.
  • Early detection of lymph node metastasis is challenging.
  • Station 8 lymph nodes are accessible during pancreatoduodenectomy (PD).

Purpose of the Study:

  • To assess if microRNA (miRNA) levels in station 8 lymph nodes correlate with postresection recurrence in PAC.
  • To evaluate specific miRNAs (miRNA-10b, miRNA-30c, miRNA-21, miRNA-155) and CK19 mRNA expression.

Main Methods:

  • Quantitative PCR was used to assay miRNA and CK19 mRNA levels in station 8 lymph nodes from 37 patients.
  • Expression levels were correlated with disease recurrence, recurrence-free survival (RFS), and overall survival (OS).

Main Results:

  • Increased miRNA-10b levels were observed in 14/30 PAC patients.
  • High miRNA-10b correlated with earlier cancer recurrence and shorter RFS (42.5 vs. 92.4 weeks).
  • Other tested miRNAs and CK19 mRNA did not correlate with RFS or OS.

Conclusions:

  • Elevated miRNA-10b in station 8 lymph nodes may serve as a biomarker for early disease progression in periampullary tumors.
  • This could help assess recurrence risk in patients undergoing PD.
