Tumor necrosis factor (TNF) may drive chronic lymphocytic leukemia (CLL) progression. Early-stage CLL patients showed higher TNF production, suggesting its role in the disease.

- Stimulated cells with phytohemagglutinin (PHA) and bacterial lipopolysaccharide (LPS).
- Measured TNFα cytotoxic activity in conditioned media from stimulated and unstimulated cells and compared to normal PBMC controls.

- Peripheral blood mononuclear cells (PBMC) from stage IV CLL patients exhibited weak or absent responses to stimulation.
- Stimulation of stage 0 CLL PBMC induced significantly increased TNFα production.
- Unstimulated stage 0 PBMC and B cells spontaneously released excess TNFα compared to normal controls.
- No detectable TNFα activity was observed in unstimulated peripheral blood mononuclear cells (PBMC) or B cells from stage IV CLL patients.
Conclusions:
- These findings suggest a potential role for TNFα in the progression of chronic lymphocytic leukemia (CLL).
- Differential TNFα production based on disease stage indicates its involvement in CLL pathogenesis.
