Role Of Notch Signalling In Intestinal Stem Cell Renewal01:12

Role Of Notch Signalling In Intestinal Stem Cell Renewal

2.6K
Notch signaling was first discovered in Drosophila melanogaster, where it is involved in cell lineage differentiation. Notch signaling regulates the maintenance and differentiation of intestinal stem cells or ISCs by controlling the expression of atonal homolog 1 or Atoh1. Atoh1 directs cells to differentiate into secretory cells.
Direct cell-to-cell contact is needed for the activation of Notch signaling. The signal is initiated when a notch ligand binds to a receptor on an adjacent cell, also...

Notch Signaling Pathway03:14

Notch Signaling Pathway

6.8K
The Notch signaling pathway is a major intracellular signaling pathway that is highly conserved over a broad spectrum of metazoan species. It stands unique from other intracellular signaling mechanisms in animals because notch protein itself acts as the receptor as well as the primary signaling molecule.
The Notch gene came into the limelight in 1914 after the discovery that its mutation in Drosophila melanogaster leads to a serrated (or "notched") wing margin phenotype. It was not...

Summary

Notch1 signaling drives gastric cancer stemness by upregulating CD133 expression in diffuse-type cancers. Inhibiting Notch1 may offer a targeted therapy for CD133-positive gastric tumors.

Area of Science:

  • Oncology
  • Molecular Biology
  • Cancer Stem Cell Research

Background:

  • CD133 is a stem-like cell marker in gastric cancers.
  • Notch1 signaling is crucial for stem cell maintenance and differentiation.

Purpose of the Study:

  • Investigate Notch1's role in gastric cancer development.
  • Determine if Notch1 signaling induces CD133 expression.

Main Methods:

  • Analyzed CD133 expression in gastric cancer subtypes.
  • Utilized cell lines with varying CD133 and Notch1 expression.
  • Inhibited Notch1 signaling and assessed effects on CD133, stem cell markers, and proliferation.
  • Performed chromatin immunoprecipitation and RBP-Jκ knockdown experiments.

Main Results:

  • CD133 was present in 51.4% of diffuse-type gastric cancers, not intestinal types.
  • Poorly differentiated gastric cancer cell lines showed CD133 and activated Notch1.
  • Notch1 inhibition reduced CD133, side population cells, proliferation, and anchorage-independent growth.
  • Activated Notch1 directly binds to the CD133 promoter via RBP-Jκ.

Conclusions:

  • Notch1 signaling maintains the cancer stem-like phenotype in diffuse-type gastric cancer via RBP-Jκ.
  • Targeting Notch1 signaling could be a therapeutic strategy for CD133-positive diffuse-type gastric cancers.
