Summary

The EndoKer method accurately analyzes corneal endothelium cell mosaics using slit-lamp biomicroscope images. This fully automatic approach offers a viable alternative to dedicated instruments, with results comparable to manual counting.

Area of Science:

  • Ophthalmology
  • Biomedical Engineering
  • Image Analysis

Background:

  • Corneal endothelium morphometric analysis is crucial for assessing ocular health.
  • Existing methods may have limitations in terms of area coverage and automation.

Purpose of the Study:

  • To evaluate the accuracy of the EndoKer method for automated morphometric analysis of the corneal endothelium.
  • To compare EndoKer results with manual cell counting and a dedicated microscope.

Main Methods:

  • EndoKer software was used for fully automatic cell recognition on slit-lamp biomicroscope images.
  • Analyses included in vitro (polystyrene beads) and in vivo (30 adults) datasets.
  • Accuracy was assessed by comparing EndoKer with manual cell counts and Tomey EM3000 microscope data.

Main Results:

  • EndoKer demonstrated high accuracy, with only a few percent difference in cell density and hexagonality compared to manual counts.
  • The accuracy is attributed to high-resolution slit-lamp images and improved automatic cell recognition.
  • Good agreement was observed between EndoKer and Tomey EM3000 microscope results.

Conclusions:

  • Slit-lamp biomicroscopy with EndoKer is a potentially viable alternative to dedicated endothelial instruments.
  • This method offers advantages such as a larger investigated area and imaging flexibility.
  • The method is pre-calibrated, simplifying user operation.
