Background:
- Glucocorticoid-induced osteoporosis (GIO) increases bone marrow fat, shifting mesenchymal stem cell (MSC) differentiation towards fat (adipogenesis) instead of bone (osteoblastogenesis).
- Vanadate, a protein tyrosine phosphatase inhibitor, previously prevented GIO in rats, but its direct effect on bone-derived MSCs was unknown.
Purpose of the Study:
- To investigate the effect of vanadate on adipogenesis in rat bone marrow MSCs (bmMSCs) and proximal femur MSCs (pfMSCs).
- To characterize differences in adipogenic and osteogenic potential between bmMSCs and pfMSCs.

- Vanadate effectively inhibits adipogenesis in bone-derived MSCs, offering a potential therapeutic strategy for GIO.
- Increased bone marrow adiposity in GIO may involve enhanced adipogenesis in MSCs outside the bone marrow niche, not solely direct glucocorticoid effects.
