Multidrug resistance in Kluyveromyces lactis involves efflux pumps regulated by KlPdr1p. This study identified KlPDR1 targets and explored its interaction with KlYap1p, revealing their roles in drug resistance.
Area of Science:
- Microbiology
- Molecular Biology
- Biochemistry
Background:
- Efflux pump overexpression drives multidrug resistance.
- KlPdr1p is a key regulator of efflux pump expression in Kluyveromyces lactis.
Purpose of the Study:
- Investigate KlPDR1-mediated drug resistance mechanisms in K. lactis.
- Identify genes regulated by KlPDR1.
- Examine the interplay between KlPdr1p and KlYap1p transcription factors.
Main Methods:
- DNA microarrays were used to analyze gene expression changes.
- Gene expression was studied in KlPDR1 deletion mutants and overexpressing strains.
- The role of KlYap1p in KlPDR1 regulation was assessed.
Main Results:
- Eighty-nine KlPDR1 targets were identified.
- 16 genes were consistently induced by KlPDR1 overexpression and repressed by KlPDR1 deletion.
- KlPDR1 expression increased with hydrogen peroxide and depended on KlYap1p, but KlYap1p did not regulate KlPDR1 with azoles.
Conclusions:
- KlPdr1p and KlYap1p are involved in K. lactis multidrug resistance.
- Further research is required to elucidate the precise interplay between KlPdr1p and KlYap1p.
