Summary

Aging muscle shows increased denervated fibers due to failed reinnervation. This failure may stem from a limited neurotrophin response, hindering axonal sprouting and contributing to muscle atrophy in older individuals.

Area of Science:

  • Muscle physiology
  • Neuroscience
  • Aging research

Background:

  • Skeletal muscle accumulates denervated myofibers with age, accelerating muscle atrophy.
  • Understanding the mechanisms of failed reinnervation in aging muscle is crucial.

Purpose of the Study:

  • To test if aging-related denervated myofiber accumulation results from failed reinnervation.
  • To compare reinnervation capacity in aging rats and a mouse model of denervation.

Main Methods:

  • Examined muscle from young adult and very old rats.
  • Utilized a murine model of sporadic denervation (Sarco mouse).
  • Analyzed fiber-type grouping, endplate markers, and gene/miRNA expression related to denervation and reinnervation.

Main Results:

  • Aging rat and Sarco mouse muscles showed fiber-type grouping, indicating denervation/reinnervation cycles.
  • Very old rat muscle exhibited persistent denervation with upregulated denervation transcripts and increased rapsyn.
  • Aging muscle showed a limited neurotrophin response and elevated suppressive miRNAs, suggesting impaired axonal sprouting.

Conclusions:

  • Failed reinnervation contributes to denervated fiber accumulation in aging muscle.
  • Impaired neurotrophin response and axonal sprouting may exacerbate denervation in aging muscle.
