Background:

  • Plasmacytomas (MPC) are B-cell tumors often induced by mineral oil (pristane).
  • Radiochimeras (RCh) allow for tracking of tumor cell origin.
  • Abelson virus is known to induce lymphoid tumors.

Purpose of the Study:

  • To investigate MPC induction in BALB/c radiochimeras (RCh).
  • To characterize the role of pristane and Abelson virus in MPC development.
  • To identify donor vs. host genetic factors influencing tumor susceptibility.

Main Methods:

  • BALB/c mice were rendered radiochimeras and repopulated with syngeneic hemopoietic cells.
  • Groups received either pristane followed by Abelson virus, or multiple pristane injections.
  • Tumor incidence, latency, and origin (donor vs. host) were analyzed.

Main Results:

  • Pristane + Abelson virus induced MPC in 18.5% of RCh with a short latency (83 days).
  • Multiple pristane injections resulted in a low MPC incidence (2%), with most tumors of host origin.
  • Pristane-treated RCh showed reduced oil granuloma (OG) formation compared to intact mice.

Conclusions:

  • Abelson virus can effectively induce MPC in RCh, overcoming reduced OG formation.
  • This model system is valuable for studying MPC precursor cells.
  • It facilitates the investigation of genetic factors influencing tumor development at the donor and host level.
