Background:

  • Raynaud's phenomenon is characterized by exaggerated vasoconstriction of peripheral arteries.
  • The role of platelet activation and aggregation in Raynaud's pathophysiology remains incompletely understood.

Purpose of the Study:

  • To investigate in vitro platelet aggregation responses in patients with primary and secondary Raynaud's phenomenon.
  • To compare platelet reactivity to various aggregating agents between patients and healthy controls.

Main Methods:

  • Platelet aggregation was assessed in vitro using serotonin (5-HT), adenosine diphosphate, adrenaline, and collagen.
  • 16 patients with Raynaud's phenomenon and 13 healthy volunteers were included in the study.

Main Results:

  • Platelets from Raynaud's patients showed significantly enhanced aggregation to serotonin (5-HT) and low-dose adenosine diphosphate.
  • Responses to collagen and adrenaline were normal in patients.
  • Secondary Raynaud's patients displayed greater adenosine diphosphate hypersensitivity than primary Raynaud's patients.

Conclusions:

  • Enhanced platelet reactivity to 5-HT and adenosine diphosphate in Raynaud's phenomenon supports a role for these agents in the disease.
  • The findings suggest potential therapeutic targets for managing Raynaud's phenomenon.
