DNA Microarrays02:34

DNA Microarrays

21.9K
Microarrays are high-throughput and relatively inexpensive assays that can be automated to analyze large quantities of data at a time. They are used in genome-wide studies to compare gene or protein expression under two varied conditions, such as healthy and diseased states. Microarrays consist of glass or silica slides on which probe molecules are covalently attached through surface functionalization. Most commonly, the slides are prepared through the chemisorption of silanes to silica...

Ribosome Profiling02:24

Ribosome Profiling

4.3K
Ribosome profiling or ribo-sequencing is a deep sequencing technique that produces a snapshot of active translation in a cell. It selectively sequences the mRNAs protected by ribosomes to get an insight into a cell’s translation landscape at any given point in time.
Applications of ribosome profiling
Ribosome profiling has many applications, including in vivo monitoring of translation inside a particular organ or tissue type and quantifying new protein synthesis levels.
The technique...

Summary
This summary is machine-generated.

Long non-coding RNAs (lncRNAs) show altered expression in dermatomyositis (DM) muscle. Aberrant lncRNAs and mRNAs, particularly involving type 1 interferon signaling, offer insights into DM pathogenesis.

Area of Science:

  • Genomics
  • Immunology
  • Molecular Biology

Background:

  • Long non-coding RNAs (lncRNAs) are functional genomic transcripts with largely unknown roles in autoimmune diseases.
  • Dermatomyositis (DM) is an idiopathic inflammatory myopathy with complex pathogenesis.
  • The involvement of lncRNAs in DM muscle tissue has not been previously investigated.

Purpose of the Study:

  • To profile and identify differentially expressed lncRNAs and mRNAs in the muscle tissue of patients with dermatomyositis.
  • To explore the potential roles of these non-coding RNAs in the pathogenesis of DM.
  • To investigate the association of lncRNA expression patterns with clinical subgroups of DM.

Main Methods:

  • A lncRNA + mRNA microarray analysis was conducted on muscle tissue from 15 treatment-naive DM patients and 5 healthy controls.
  • Bioinformatic analyses, including pathway and gene ontology analysis, were performed on differentially expressed genes.
  • Bioinformatics prediction was used to identify potential regulatory interactions between lncRNAs and target genes.

Main Results:

  • A total of 1198 lncRNAs and 1213 mRNAs were found to be significantly differentially expressed in DM patients compared to controls.
  • Type 1 interferon signaling was identified as a significantly dysregulated pathway in all DM subgroups.
  • Specific lncRNAs, such as linc-DGCR6-1, were predicted to regulate key genes like USP18, which is highly expressed in DM muscle fibers.

Conclusions:

  • This study provides the first comprehensive overview of aberrantly expressed lncRNAs in DM muscle tissue.
  • The findings highlight the dysregulation of type 1 interferon signaling and identify potential lncRNA targets involved in DM pathogenesis.
  • The results expand the understanding of molecular mechanisms underlying dermatomyositis and suggest novel avenues for research.
