Bioavailability Enhancement: Drug Stability Enhancement and GI Retention

292
Improving a drug's stability in the gastrointestinal (GI) tract is paramount for enhancing its bioavailability and therapeutic effectiveness. Various strategies are employed to protect the drug from the harsh gastric milieu and to ensure its release and absorption at the desired site within the GI tract.Polymer coatings are one such method used to shield drugs from the stomach's acidic environment. By preventing premature drug release, these coatings improve the bioavailability of unstable...

Oral Drug Delivery Systems: Continuous-Release Systems

135
Continuous-release drug delivery systems offer a strategic approach to maintaining therapeutic drug levels over extended periods following oral administration. By modulating the release rate of active pharmaceutical ingredients, these systems minimize fluctuations in plasma concentrations, which enhances clinical efficacy and reduces the need for frequent dosing. Such characteristics make them particularly advantageous in managing chronic diseases where patient adherence and stable drug...

Area of Science:

  • Materials Science
  • Pharmaceutical Science
  • Biochemistry

Background:

  • Chitosan's antioxidant properties are being explored for pharmaceutical applications.
  • Low molecular weight (LMW) chitosans (CS1-CS4) with varying molecular weights (22-81 kDa) were investigated.
  • Oxidative stress is a significant factor in various health conditions.

Purpose of the Study:

  • To evaluate the antioxidant capacity of LMW chitosans for potential use in extended-release tablets.
  • To assess the drug release profile and antioxidant activity of chitosan-based extended-release theophylline tablets.

Main Methods:

  • Antioxidant activity assessed via reduction of Cu2+, hydroxyl, superoxide, and DPPH radicals using ESR spectrometry.
  • Extended-release tablets formulated using spray drying with theophylline as a model drug.
  • Drug release kinetics and antioxidant activity (ABTS radical scavenging) of formulated tablets were evaluated.

Main Results:

  • CS2 (38 kDa) exhibited the highest radical scavenging activity among the tested chitosans.
  • Theophylline release from chitosan tablets increased with increasing chitosan molecular weight.
  • CS2-theophylline tablets demonstrated superior antioxidant activity compared to other chitosan formulations.

Conclusions:

  • CS2 possesses significant antioxidant properties, indicating its potential for combating oxidative stress.
  • CS2-based extended-release theophylline tablets show promise as a dual-action formulation with high antioxidant capacity.
