In multiple sclerosis (MS) patients, early B cell development in bone marrow appears normal. However, a reduced proportion of natural killer T cells was observed in the bone marrow of MS patients.
Area of Science:
- Immunology
- Neurology
- Hematology
Background:
- Multiple sclerosis (MS) immunopathology is thought to originate in the peripheral lymphatic system.
- The precise location and nature of the initiating events in MS remain unclear.
Purpose of the Study:
- To investigate early hematopoiesis in the bone marrow of MS patients.
- To compare the proportions of specific immune cell subsets in the bone marrow of MS patients and healthy controls.
Main Methods:
- Flow cytometry (FACS) analysis was used to examine bone marrow cell subsets.
- The study included 9 MS patients and 11 neurologically healthy controls.
Main Results:
- A statistically significant lower proportion of natural killer T cells (P=0.045) was found in the bone marrow of MS patients compared to controls.
- Proportions of hematogenous stem cells, myeloblasts, and B cell precursor subsets did not differ between the two groups.
Conclusions:
- This pilot study suggests that early B cell lineage development in the bone marrow is not significantly altered in MS patients.
- Further research is needed to elucidate the role of natural killer T cells in MS pathogenesis.
