Summary

Researchers identified a newt pax-6 gene crucial for eye development. Sonic hedgehog (shh) RNA inhibited pax-6 expression, impacting eye cup formation, but not in surface ectoderm.

Area of Science:

  • Developmental Biology
  • Genetics
  • Molecular Biology

Background:

  • Pax-6 is a conserved master control gene essential for eye development across diverse species.
  • Understanding the regulation of pax-6 is key to deciphering the intricate mechanisms of eye formation.

Purpose of the Study:

  • To isolate and characterize the newt (Cynops pyrrhogaster) pax-6 gene.
  • To investigate the role of sonic hedgehog (shh) in regulating pax-6 expression during eye development.
  • To determine the earliest stage of ectodermal competence for lens induction.

Main Methods:

  • Isolation of newt pax-6 cDNA using RT-PCR and sequence analysis.
  • Analysis of alternative splicing variants of the newt pax-6 gene.
  • Expression pattern analysis of pax-6 in newt embryos.
  • In vivo and in vitro experiments involving sonic hedgehog (shh) RNA injection and animal cap assays.

Main Results:

  • Four alternatively spliced forms of the newt pax-6 gene were identified.
  • Newt pax-6 exhibited conserved sequence identity and expression patterns compared to mouse and zebrafish pax-6.
  • Pax-6 expression was observed in the neural plate, eye, nasal primordium, and central nervous system (excluding midbrain).
  • Sonic hedgehog (shh) RNA injection inhibited pax-6 expression in the optic vesicle and disrupted eye cup formation.
  • Shh did not affect pax-6 expression in the surface ectoderm overlying the optic cup.
  • Transient pax-6 activation was observed in early gastrula stage animal cap explants.

Conclusions:

  • The newt pax-6 gene is a functional homolog of pax-6 involved in eye development.
  • Sonic hedgehog (shh) plays a regulatory role in pax-6 expression within the optic vesicle but not in the overlying surface ectoderm.
  • Ectoderm may acquire competence for lens induction as early as the mid-gastrula stage.
