Synthesis and Regulation of Thyroid Hormones01:20

Synthesis and Regulation of Thyroid Hormones

8.6K
Low blood levels of the thyroid hormones — triiodothyronine (T3) and thyroxine (T4) — signal the hypothalamus to release the thyrotropin-releasing hormone (TRH). TRH then reaches the pituitary gland and stimulates the release of thyroid-stimulating hormone(TSH) into the bloodstream.
Upon reaching the thyroid gland, TSH stimulates the follicular cells' active uptake of iodide ions from the blood. The ions diffuse to the apical surface of the cells and are oxidized to iodine. The...

The Thyroid Gland01:23

The Thyroid Gland

8.3K
The thyroid gland is a small, butterfly-shaped gland located in the neck and covers the anterior surface of the trachea. The gland has two lateral lobes connected by a thin tissue mass called the isthmus. Internally, each lobe comprises many small spherical structures known as thyroid follicles, surrounded by a network of blood vessels.
The follicles have a central cavity lined by simple cuboidal to squamous epithelial cells called follicular cells. These cells produce the glycoprotein...

Functions of Thyroid Hormones

6.3K
The thyroid hormone (TH) plays a pivotal role in the intricate orchestration of physiological processes, exerting profound effects on development, metabolism, and homeostasis throughout different life stages.
TH is indispensable for the normal development and maturation of the skeletal, muscular, and nervous systems during fetal and childhood growth. It facilitates bone mineral turnover and regulates protein synthesis in developing tissues, contributing significantly to overall growth and...

Summary

We developed multianalyte immunoassays (MAIA) for thyroid function tests, including thyroxine (T4), thyroid stimulating hormone (TSH), and thyroglobulin (Tg). This sensitive and precise assay shows excellent correlation with existing methods.

Area of Science:

  • Biochemistry
  • Analytical Chemistry
  • Immunology

Background:

  • Thyroid function assessment relies on measuring key analytes like thyroxine (T4), thyroid stimulating hormone (TSH), and thyroglobulin (Tg).
  • Existing immunoassays often measure these analytes individually, necessitating multiple tests.
  • Developing a simultaneous assay for these diverse analytes is crucial for efficient thyroid diagnostics.

Purpose of the Study:

  • To develop and validate a multianalyte immunoassay (MAIA) platform for simultaneous measurement of T4, TSH, and Tg.
  • To assess the sensitivity, precision, and clinical utility of the developed MAIA.
  • To demonstrate the adaptability of the MAIA platform for both isotopic and non-isotopic tracers.

Main Methods:

  • Utilized polycarbonate (PC) track-etched membranes (TEM) as a solid support for analyte immobilization.
  • Developed competitive immunoassays for T4 and non-competitive immunoassays for TSH and Tg on the same TEM.
  • Employed Iodine-125 (125I) as a radiotracer for assay development and validation.

Main Results:

  • The developed MAIA demonstrated high sensitivity and precision for all three analytes (T4, TSH, Tg).
  • The assay exhibited a clinically useful working range, suitable for diagnostic applications.
  • Results from the MAIA correlated very well with established individual analyte immunoassays.
  • The assay format proved effective with a radiotracer and is adaptable to non-isotopic tracers.

Conclusions:

  • The developed multianalyte immunoassay (MAIA) provides a sensitive, precise, and efficient method for assessing thyroid function.
  • This platform enables simultaneous measurement of T4, TSH, and Tg, offering a potential advantage over single-analyte assays.
  • The MAIA technology is versatile and can be adapted for various detection systems, including non-isotopic methods.
