Sortilin expression and its derived peptide (PE) production are linked during adipocyte differentiation and insulin resistance. Spadin, a PE analogue, showed no impact on adipocyte function, suggesting no side effects for antidepressant use.
Area of Science:
- Biochemistry
- Cell Biology
- Metabolism
Background:
- Sortilin is a protein involved in cellular processes.
- Sortilin-derived propeptide (PE) is a related molecule.
- Spadin, a PE analogue, has antidepressant properties.
Purpose of the Study:
- To correlate sortilin expression with PE production during adipocyte differentiation, insulin resistance, and inflammation.
- To investigate the effect of spadin on adipocyte functions.
Main Methods:
- Quantitative PCR and Western-blot for sortilin mRNA and protein.
- Specific dosing method for PE quantification.
- Immunostaining for co-localization studies.
- Glucose uptake and signaling experiments with spadin.
Main Results:
- PE production correlated with sortilin expression during adipogenesis.
- TNFα-induced insulin resistance decreased sortilin and PE levels.
- Dexamethasone-induced insulin resistance did not affect sortilin or PE.
- Spadin had no measurable effect on adipocyte glucose uptake or signaling.
Conclusions:
- Sortilin expression and PE production are linked in adipocytes, particularly during inflammatory conditions.
- Spadin's lack of effect on adipocytes suggests it may not interfere with adipocyte physiology.
- These findings contribute to understanding sortilin's role in metabolic health and spadin's therapeutic potential.
