A Meta-analysis of MBL2 Polymorphisms and Tuberculosis Risk

Certain variants of the MBL2 gene, specifically rs1800450 and rs1800451, appear to offer protection against tuberculosis (TB). These MBL2 gene polymorphisms may serve as valuable genetic markers for TB resistance.

Background:
- The mannose-binding lectin 2 (MBL2) gene is a key component of the innate immune system.
- Previous research on MBL2 gene variants (rs1800451, rs1800450, rs5030737, rs7096206, rs11003125, rs7095891) and their association with tuberculosis (TB) susceptibility has yielded inconsistent results.
Purpose of the Study:
- To conduct a comprehensive meta-analysis evaluating the association between MBL2 gene polymorphisms and the risk of developing TB.
- To clarify the inconsistent findings regarding MBL2 variants and TB susceptibility.

- A meta-analysis was performed, pooling data from 22 studies involving 7095 TB patients and 7662 controls.
- Data were analyzed for associations between MBL2 alleles, genotypes, and minor allele carriers with TB risk.
- Stratified analyses were conducted based on ethnicity.

- The homozygous variant genotype (CC) of MBL2 rs1800451 was significantly associated with a reduced risk of TB (OR=0.834, p=0.045).
- No significant associations were found for other genetic models of the studied MBL2 polymorphisms (rs1800450, rs5030737, rs7096206, rs11003125, rs7095891) and combined variants.
- Stratified analysis revealed a decreased TB risk in the African population for MBL2 rs1800450 and rs1800451.
Conclusions:
- MBL2 gene polymorphisms rs1800450 and rs1800451 demonstrate a protective effect against TB infection.
- These specific MBL2 polymorphisms are significant potential genetic markers for TB resistance.
