Purpose of the Study:

  • To investigate the effects of melamine on testosterone synthesis in mice.
  • To elucidate the mechanisms underlying melamine-induced testicular damage.

Main Methods:

  • Mice were orally administered melamine (2, 10, or 50 mg/kg) for 28 days.
  • Evaluated testicular structure, sperm parameters, and serum/testicular testosterone levels.
  • Assessed expression of steroidogenic enzymes (StAR, P450scc, 17β-HSD) and Leydig cell count.

Main Results:

  • Melamine induced seminiferous tubule damage, germ cell necrosis, and sperm abnormalities.
  • A dose-dependent decrease in serum testosterone and testicular StAR, P450scc, and 17β-HSD levels was observed.
  • Melamine exposure reduced Leydig cell numbers.

Conclusions:

  • Melamine reduces testosterone levels by down-regulating steroidogenic enzyme expression and decreasing Leydig cell count.
  • These effects contribute to melamine-induced testicular damage and impaired spermatogenesis.
  • Melamine poses a risk to male reproductive health.
